I really like the new Cards skin. Quite a lot actually.
However I find the vertical scroll bar too thin and a bit hard to grab. Not impossible but tricky. (Also, a bit hard to differentiate against the dark background.)
Would it be possible to make that scroll bar a bit wider?

I would have thought the scroll bar width is set programmatically. How can I modify with image editing skills?

One place I'd start is with the Scrollbar_HorizontalBackground.png, Scrollbar_HorizontalHandle.png, Scrollbar_VerticalBackground.png and Scrollbar_VerticalHandle.png files in the skin's main directory.
But yeah, the scrollbars are done via PNG images set in the main.xml file found in the specific skin's directory.

You'll also have to edit the 2x versions of those files too if you want scaling support (past 100%).
I also recommend forking the skin itself and editing the main.xml file to reflect the fork. Or else with every new MC update, the edited files will get overridden by the originals.

It's quite horrible in a desktop environment though. It's more suited to a touchscreen set up. The scrollbars stay small untill you touch them, then they 'grow'...

That's what they are supposed to do, to make it easier to grab them. And it's optional.
As I have aged (beyond belief), I find that my hands are no longer steady and some of the fine motor skills necessary to use a mouse are gone. The fat scrollbars help a lot, even on a desktop. On a touchscreen, they are essential.
Maybe it's time for you to get a touchscreen?

Open options, type "over" (without quotes) in the search field at the bottom, select "Tree and View" from the list on the left.

Correct me if I am wrong but using the option for larger scroll bars only makes them grow when you have put your mouse over the thin scroll bar, so it is only a visual cue you have succeeded in selecting the thin bar it does not make the target you trying to hit any bigger.

With a mouse, hovering over the scroll bar enlarges it and the new, wider bar is functional.
If that isn't happening, please explain what does happen in more detail.

I think he is just saying. You still need to be over the thin bar for them to be thick. Removing some of the point of having a thinker bar.
Is there any reason to not have an option to always be thick?
