Pretty sure this can't happen automatically.  What you could do is create a smartlist that shows all 'podcast' files with a playcount of 1 or more.  Then just delete the files (from both HD and library) from inside that list.

What I do is add the 'Number Plays' field to my view for podcasts and use that to track which ones I've heard and can delete.
