Just got my first 500g HD to replace 2-200gers. Should I go for one large partition or make multiple partitions? The largest directory is about 220gigs, 4 others averaging about 35g each.

for me it would depend on what is in those directory's. i have for instance one disk where i made a big partition for my media files. but a smaller for recording music. the last one needs to be defragmented fairly often. a seperate partition makes that less time consuming.
on an other disc i have pictures and music files. i made that one partition. dont have to look if one is getting to full (10% free space), they dont bite each other and are both managed by MC. i also have one disk with all my documents, my work disc. ill keep that seperated form my media files. just because it makes me feel secure.
no sience, just my reconsiderations about this.

It's all media storage, recording and other processes are done on a different drive.

In general you should leave as a single disk.
It avoids problems later whereby you have space left on one partition but need it on the other etc.
A single drive is alot cleaner and if organised properly works great.
No real gain from partitioning a drive.

Thanx guys!

I agree.
Unless you are trying to solve a specific issue which is driving you to partition a drive, leave it as a single drive.

I recommend having a separate partition for the OS and applications (about 30GB usually depending on your requirements). Makes restoring your OS much easier, should you need to. I generally restore my OS every few months from a backup to keep the thing from slowing down due to the ghosts of uninstalled apps.

What I do too - always have the OS on a seperate partition of around this size!
