I can go to Label Editor and make changes to layout, font, etc.,
and save the file under some name I specify with an .mjl extension,
but I can't figure out how to get MJ to USE that edited layout when
I subsequently want to print using my layout. Help file doesn't
contain this info, and intuition (at least mine) didn't work either.
Any help would be appreciated. Thanks.

If MJ is open, generally you have to select some songs to open CD Labler. Then select "Go To Label Editor"
If you go to Programs and select Media Jukebox than select CD Labeler, it will open directly to Label Editor.
Select File>open look for the file name you used to save it. Then click on it.

Yes, I did get that far, but I was hoping that the changes I made
(e.g., to the font used for printing track listings) could be
saved as a template and automatically applied to future invocations
of CD Labeler for different CDs. But it appears that each CD label
is saved as its own file, and I need to edit each one individually.
That's workable, although a future enhancement that allowed the
creation of templates would be nice.

That option has been requested and is on the wish list for a future version.
