Recently when trying to open .vob files I have been getting the error:

Something went wrong with plyback. 
Details: Failed to open or parse file.  (hr=0x80004055)

I had experienced this a while ago, changed Update Channels = Latest and this fixed it until now.

I'm running 16.0.181.

Options - File Types DVD Video Options:
- Playback Method = Automatic
- Audio Decoders = Automatic

General Video Settings:
- DirectShow selection method: Red October Standard
- Video Renderer: Enhance Video Renderer

Ideas?

Thie error means DVD Navigator could not open the file. Make sure the path and permissions are correct. If they are, you should try to reimport the file(s). And/or Backup your Library, then clear it and then reimport the files.

I can play the file(s) from VLC and Media PLayer Classic so I don't see it would be a permission issue.

I'm not trying to import this into my library, I just double click the file and expect the default player (MC16) to be able to play it just like the previously mentioned players can do.  Perhaps MC tries to import it into the library through this process and is stuffing up the playing.  But even so, this isn't nice behaviour. 

A backup/clear/reimport of my entire library seems as bit excessive just to get .vob files to play.  If this is a requirement to get something to play then surely this is a bug?

My feeling is there is a deeper issue here or a setting that I have neglected to flick on/off.

It's not normal behavior for MC. That's why I think something got messed up on your system. My suggestion was something that was suggested in another post on this subject a while back - and seemed to work for that person (tip - use the Search button).
I assume the file is in your Library now. That is why I suggested re-importing it. Maybe the first time you added it the path got messed up. If it is not in your Library, where is it?

To test playing movies outsdie the Library, I just popped in a DVD disk and selected one of the VOB files (from Windows Exlporer) and it opened and played fine. So I don't think there is a bug in MC.

In the MC settings (Tools>Options>File Types do you have the "File Association" box "checked" for DVD Video (vob, ifo) files? Do you have the Playback method set to Automatic or something else?

I am having the exact same issue.  File is saved on my drive and not added to the library.

Playback is set to automatic. File is associated with MC16. It does not work when I double click a vob file in Windows Explorer.

Does not make sense why it stopped working.
