First, let me say that I LOVE the new View Filter icon on the toolbar. This makes the Media Library sooo much more useful.

However, it really needs a simple improvement: Allow arbitrary search strings for Include and Exclude.  Right now, it limits you to choosing a single playlist for each of these, which means that every time I want filter on an arbitrary search criteria, I have to temporarily create a smartlist first.

For UI, you could consider putting a textbox directly in the pulldown menu, just like Microsoft Access does, for its filtering.
