The usual way is the play them is like any other video file (e.g. Import > Select > Play), but I am guessing that you have some kind of playback problem with your Matroska file or files.
Without detailed information it is difficult to answer, but many Matroska problems have been resolved by installing additional DirectShow filters (the Windows DirectShow playback system does not have native native support for MKV files). You could install the CCCP filter and codec pack:

- Problem description. What do you try to do and what does happen then?
- Does the problem occur only with certain files? Do some other Matroska files play correctly? Do other video file formats play correctly?
- The exact file format. Matroska is a container format and it can contain many kinds of file data. You can inspect video files with the MediaInfo tool:
