eSATA would be much better than USB 2, USB 3.0, but not better than USB 3.1 Gen 2. So it depends on the standard the enclosure uses. But if you are only accessing one drive in the enclosure at a time, the USB 2 at 480 Mbps or about 60 MB/s should be okay. So playback or copying a set of files to one drive at a time wouldn't benefit much from a faster connection.
Your graphics benchmark is insanely low. Are you on Windows 10? There have been several Windows 10 updates that have broken things, and one recent one forced me to update graphics drivers, because of some introduced incompatibility. So I would be upgrading graphics drivers rather than reinstalling MC or anything similar.
Also if you have upgraded to the lastest MC23 and are using Red October High Quality, then you are now using a new version of madVR. If you have customised any settings for madVR previously then they may be causing some problems. Note your customisations, but then reset madVR to the defaults. See if that improves things.
